Contribution à l'étude de vibrions pathogènes d'huîtres creuses Crassostrea gigas - Interactions hémocytes-vibrions et développement de tests de diagnostic d'intérêt taxonomique ArchiMer

Les bactéries du genre Vibrio représentent le groupe le plus fréquemment associé à des maladies infectieuses chez les mollusques marins et notamment l'huître creuse Crassostrea gigas. A ce jour trois espèces de Vibrio pathogènes d'huîtres creuses V. splendidus, V. aestuarianus et récemment V. coralliitycus ont été identifiés en France. La prévention des mortalités associées à ces vibrioses repose sur une meilleure compréhension des interactions hôte-pathogène et sur l'utilisation de techniques de diagnostic rapides à mettre en oeuvre et fiables. Vibrio splendidus LGP32 is a bacterial pathogen associated to the summer mortality outbreaks that have affected the production of Crassostrea gigas oysters over the past decades. We showed here that the OmpU porin is a major effector of the V. splendidus / C. gigas interaction. For that, we have constructed a ΔompU mutant of V. splendidus, and shown that the OmpU porin is implicated (i) in the resistance of V. splendidus to antimicrobials, including those of oyster, (ii) in its in vivo fitness, and (iii) in its virulence in oyster experimental infections (mortalities have been reduced from 56 % to 11 % upon mutation). Rearing of Crassostrea gigas is the most economically important aquaculture activity in France. Bacteria belonging to Vibrio genus constitute one of the most abundant bacterial group in marine ecosystems. Two Vibrio species, V. splendidus and V. aestuarianus, were associated with many cases of mortality events in reared C. gigas spat and juvenile oysters, commonly during summer. Outer membrane vesicles are vehicles for the delivery of Vibrio tasmaniensis virulence factors to oyster immune cells ArchiMer

Vibrio tasmaniensis LGP32, a facultative intracellular pathogen of oyster haemocytes, was shown here to release outer membrane vesicles (OMVs) both in the extracellular milieu and inside haemocytes. Intracellular release of OMVs occurred inside phagosomes of intact haemocytes having phagocytosed few vibrios as well as in damaged haemocytes containing large vacuoles heavily loaded with LGP32. The OMV proteome of LGP32 was shown to be rich in hydrolases (25%) including potential virulence factors such as proteases, lipases, phospholipases, haemolysins and nucleases. Resistance to Antimicrobial Peptides in Vibrios ArchiMer

Vibrios are associated with a broad diversity of hosts that produce antimicrobial peptides (AMPs) as part of their defense against microbial infections. In particular, vibrios colonize epithelia, which function as protective barriers and express AMPs as a first line of chemical defense against pathogens. Recent studies have shown they can also colonize phagocytes, key components of the animal immune system. Phagocytes infiltrate infected tissues and use AMPs to kill the phagocytosed microorganisms intracellularly, or deliver their antimicrobial content extracellularly to circumvent tissue infection. The major outer membrane protein OmpU of Vibrio splendidus contributes to host antimicrobial peptide resistance and is required for virulence in the oyster Crassostrea gigas ArchiMer

Vibrio splendidus, strain LGP32, is an oyster pathogen associated with the summer mortalities affecting the production of Crassostrea gigas oysters worldwide. Vibrio splendidus LGP32 was shown to resist to up to 10 mu M Cg-Def defensin and Cg-BPI bactericidal permeability increasing protein, two antimicrobial peptides/proteins (AMPs) involved in C. gigas immunity. The resistance to both oyster Cg-Def and Cg-BPI and standard AMPs (polymyxin B, protegrin, human BPI) was dependent on the ompU gene. Indeed, upon ompU inactivation, minimal bactericidal concentrations decreased by up to fourfold. AMP resistance was restored upon ectopic expression of ompU. OmpU porins are increasingly recognized as key determinants of pathogenic host Vibrio interactions. Although mechanisms remain incompletely understood, various species, including the human pathogen Vibrio cholera, require OmpU for host colonization and virulence. We have shown previously that OmpU is essential for virulence in the oyster pathogen Vibrio splendidus LGP32. Here, we showed that V. splendidus LGP32 invades the oyster immune cells, the hemocytes, through subversion of host-cell actin cytoskeleton.
